This month, we updated our PARP inhibitor records based on a new and important publication reporting potency and selectivity information for 11 PARP family inhibitors that are commonly used as chemical probes (Thorsell et al., 2016). This paper reports several key new observations about the activity of these inhibitors, prompting us to revisit and update the probe and historic compound records.

The Chemical Probes Portal is excited to announce a growth campaign focusing on the Tyrosine Kinases. This class of proteins is already known to contain important drug targets, as the class includes many growth factor receptors that drive tumor growth. Tyrosine kinases come in two basic flavors, receptor and non-receptor kinases, and not all of them have been well studied.

In this issue, we highlight how the rigorous application of chemical probes in biological systems can lead to better probes and improve drug discovery efforts through a case study of ABT-199 (venetoclax). We also highlight two programs that we rolled out late in 2016: Historic Compounds and Vendor Links.

The Portal now features a new class of compounds called Historic Compounds. These small molecules are typically non-selective or not sufficiently potent compared with other available chemical probes to merit the probe designation.
A table of all Historic Compounds currently posted to the portal is found here. To recommend we add a historic compound - use our probe submission form.

This guest post by Ellen Berg defines a new initiative for the Portal - a chemical probe collection that is suitable for characterizing phenotypic screens - and will be formalized soon as a Portal Resource that will be maintained and updated with guidance from a subcommittee of our Scientific Advisory Board.

We are pleased to announce the publication of new chemical probes in the Portal. We have recently released validation data for the probes listed below. These probes are still under review by our SAB so it will take a few weeks for ratings and recommendations to emerge. Stay tuned, as we will be releasing more probes as well as inviting new submissions soon!

At the Chemical Probes Portal, our mission is to help change the way scientists find and use chemical probes. In addition to providing expert recommendations that guide probe selection and use, we aim to help make it easier for scientists to gain access to the high quality reagents they need to perform robust and reliable experiments. Thus, we are pleased to provide links from our chemical probe pages to the pages of trusted vendors where users can obtain the chemical probes they need.
